Biological Background: ATN1 Function and Localization
- Atrophin-1 (ATN1), also known as dentatorubral-pallidoluysian atrophy protein, is a transcriptional corepressor involved in regulating gene expression.
- It recruits NR2E1 to repress transcription and functions as a corepressor for MTG8, with intrinsic repression activity independent of poly-glutamine repeat length.
- ATN1 promotes vascular smooth muscle cell migration and orientation (by similarity), suggesting a role in cardiovascular biology.
- The protein localizes to the nucleus and cytoplasm, particularly the perinuclear region, and is also found at cell junctions.
- ATN1 is widely expressed in adult tissues, with high levels in the brain, including the amygdala, caudate nucleus, and hippocampus, and elevated expression in fetal tissues.
- Post-translational modifications include acetylation, methylation, phosphorylation, and ubiquitination, which may regulate its activity and stability.
- Mutations involving poly-glutamine tract expansion cause dentatorubral-pallidoluysian atrophy (DRPLA), a neurodegenerative disorder characterized by epilepsy, intellectual disability, and ataxia.
Experimental Guidance and Technical Tips
- The immunogen covers the C-terminal region (aa 900-1040) of human ATN1, which is distal to the polyQ repeat tract; this design may help reduce interference from repeat length polymorphisms in detection.
- For Western blot, the predicted molecular weight is approximately 125 kDa; ensure adequate transfer conditions for high-molecular-weight proteins.
- For immunofluorescence, consider permeabilization protocols that allow detection of both nuclear and cytoplasmic pools.
- Validate antibody specificity in the relevant human tissue or cell line model, especially those with known ATN1 expression, such as brain or fetal tissues.
