Biological Background: ATM Function and Localization
- ATM, or Ataxia telangiectasia mutated, is a serine/threonine protein kinase that acts as a DNA damage sensor, recognizing double-strand breaks (DSBs) and activating checkpoint signaling in response to genotoxic stresses such as ionizing radiation and ultraviolet A light (PubMed:10550055, PubMed:10839545, PubMed:10910365). - The kinase phosphorylates downstream effectors containing the consensus motif [ST]-Q, including histone H2AX at Ser-139, p53/TP53, CHEK2, BRCA1, MRE11, and many others, thereby coordinating DNA repair, cell cycle arrest, and apoptosis (PubMed:10550055, PubMed:10766245, PubMed:10802669). - ATM plays a key role in cell cycle control and is considered a tumor suppressor, with mutations in the ATM gene linked to ataxia-telangiectasia, a syndrome featuring neurodegeneration, immunodeficiency, and cancer predisposition.

- For Western blotting, use appropriate gel percentage and transfer conditions (e.g., wet transfer with extended time) to efficiently blot this high-molecular-weight protein.
- In immunofluorescence/immunocytochemistry, optimize fixation and permeabilization to detect ATM in nuclear and cytoplasmic compartments; consider using known DNA damage-inducing treatments to evaluate localization changes.
