Biological Background: ATL2 Function and Localization
- ATL2, also known as ARL6IP2 or atlastin-2, is a member of the atlastin family of GTPases with a molecular weight of approximately 66 kDa.
- Mediates GTP-dependent fusion of endoplasmic reticulum (ER) membranes, maintaining the continuous ER network, and facilitates the formation of three-way junctions where ER tubules intersect. Related references: PMID:18270207, PMID:19665976, PMID:22065636
- Upon GTP hydrolysis, the homodimeric complex undergoes conformational changes through its 3HB regions, pulling opposing membranes together to drive fusion; after phosphate release, the dimer disassembles to reset the cycle.
- Localized to the endoplasmic reticulum membrane via two transmembrane helices.
- Expressed in peripheral tissues at the protein level.
- Contains a coiled-coil domain and is subject to post-translational modifications including methylation and phosphorylation; alternative splicing produces multiple isoforms.
