Biological Background: ATG7 Function and Localization

  • ATG7 is an E1-like activating enzyme essential for two ubiquitin-like conjugation systems that drive autophagy and cytoplasm-to-vacuole transport (Cvt).
  • It activates ATG12 for conjugation with ATG5, and primes ATG8 family proteins (such as LC3) for lipidation with phosphatidylethanolamine, a step critical for autophagosome membrane association.

  • For Western blot (WB), a band around 78 kDa is expected; consider loading adequate protein and optimizing transfer conditions to capture the full-length protein.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), ATG7 staining may appear diffuse in the cytoplasm with potential punctate enrichment at preautophagosomal structures under autophagy-inducing conditions. Validate with autophagy markers (e.g., LC3) in the relevant tissue or cell model.
  • For ELISA applications, the antibody may serve as a capture or detection reagent; however, pairing with a suitable matched antibody and optimizing coating conditions are advised for best performance.
