Biological Background: ATG7 Function and Localization
- E1-like activating enzyme essential for the two ubiquitin-like conjugation systems required for autophagy and cytoplasm-to-vacuole transport.
- Activates ATG12 for conjugation with ATG5, and the ATG8 family for conjugation with phosphatidylethanolamine, driving autophagosome formation.
- Facilitates LC3-I lipidation to generate LC3-II, which localizes to autophagosomal membranes. Related references: PMID:34161705
- Localizes to the cytoplasm and preautophagosomal structures, consistent with its role in early autophagy steps.
- Widely expressed across human tissues, notably enriched in kidney, liver, lymph nodes, and bone marrow.
- Regulates diverse processes including mitophagy, axonal homeostasis, hematopoietic stem cell maintenance, Paneth cell granule formation, and adipose differentiation.
- Post-translationally modified by acetylation and phosphorylation; belongs to the ubiquitin-activating E1-like protein family.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 500–676 in the C-terminal region of human ATG7; consider epitope accessibility in your assay design.
- For IHC-P, optimize antigen retrieval and antibody dilution using human tissue sections with known ATG7 expression (e.g., kidney, liver).
- In ELISA, validate coating conditions with recombinant ATG7 protein or peptide and include appropriate positive/negative controls.
- Since cross-reactivity is limited to human, verify performance in your specific sample matrix.
