Biological Background: ATG5 Function and Localization
- Autophagy protein 5 (ATG5), also known as APG5-like, ASP, or hAPG5, is a key autophagy-related protein encoded by the ATG5 gene (Gene ID: 9474) in humans.
- ATG5 is essential for autophagic vesicle formation. It conjugates with ATG12 through a ubiquitin-like system involving ATG7 (E1-like) and ATG10 (E2-like). The ATG12-ATG5 conjugate acts as an E3-like enzyme for lipidation of ATG8 family proteins, facilitating their association with vesicle membranes.
- ATG5 plays a critical role in mitochondrial quality control after oxidative damage, contributing to cellular longevity, and is involved in lymphocyte development, where it is essential for B and T cell survival and proliferation.
- The protein is required for optimal antigen processing and presentation via MHC II, maintenance of axon morphology, normal adipocyte differentiation, and promotes primary ciliogenesis by removing OFD1 from centriolar satellites and degrading IFT20 via autophagy.
- ATG5 may also function in apoptosis, possibly within the modified cytoskeleton. Its expression is upregulated in apoptotic cells downstream of caspase activity, and it is crucial for IFN-gamma-induced autophagic cell death by interacting with FADD.
- (Microbial infection) ATG5 can act as a proviral factor: it negatively regulates innate antiviral immune response by impairing type I IFN production upon VSV infection, and is required for initiation of hepatitis C virus RNA translation and replication. - Subcellularly, ATG5 localizes to the cytoplasm and the preautophagosomal structure membrane. It is ubiquitously expressed at the mRNA level, but the protein is dramatically highly expressed in apoptotic cells.

- The immunogen is a synthetic peptide within amino acids 1-100 of human ATG5. Validate specificity using ATG5 knockout cell lines or siRNA knockdown.
- For Western blot (WB) detection, a band around 32-35 kDa (ATG5 monomer; conjugated ATG12-ATG5 migrates higher at ~55 kDa) is expected. Consider testing in autophagy-induced samples (e.g., starved or rapamycin-treated) to enhance signal.
- In immunofluorescence (IF) and immunohistochemistry (IHC), ATG5 shows cytoplasmic punctate staining that accumulates upon autophagy induction; optimize fixation and permeabilization to preserve autophagosome structures.
- For immunoprecipitation (IP), the antibody may capture both free ATG5 and the ATG12-ATG5 conjugate; ensure appropriate lysis conditions to detect protein-protein interactions.
- Cross-reactivity with mouse and rat is expected based on sequence homology; nonetheless, verify performance in the relevant species and tissue context.
