Biological Background: ATG4D Function and Localization
- ATG4D is a cysteine protease (autophagin-4) belonging to the ATG4 family, which includes ATG4A, ATG4B, and ATG4C; it primarily mediates delipidation of ATG8 family proteins.
- Functions in autophagy by both proteolytic activation and delipidation of ATG8 proteins (MAP1LC3 and GABARAPL2); cleaves the C-terminal amino acid to expose a glycine necessary for conjugation to phosphatidylethanolamine (PE) and membrane insertion. Related references: PMID:21177865
- Mediates delipidation of PE-conjugated ATG8 proteins during macroautophagy and also delipidates ATG8 proteins conjugated to phosphatidylserine (PS) during non-canonical autophagy at endolysosomal compartments. Related references: PMID:29458288 PMID:33909989
