Biological Background: ATG4C Function and Localization

  • ATG4C is a cysteine protease belonging to the ATG4 family, which includes ATG4A, ATG4B, and ATG4D, with distinct substrate specificities.
  • It plays a dual role in autophagy: proteolytic activation of ATG8 family proteins (cleaving C-terminal residues to expose glycine) and delipidation of PE-conjugated ATG8 proteins. Related references: PMID:21177865 PMID:29458288 PMID:30661429
  • Compared to ATG4B, ATG4C shows weaker proteolytic activation but stronger delipidation activity. Related references: PMID:29458288
  • It localizes to the cytoplasm.
  • Post-translational modifications include acetylation and phosphorylation.
  • It is involved in the ubiquitin-like conjugation pathway essential for autophagosome formation.
  • ATG4C is weakly or not involved in phagophore growth during mitophagy. Related references: PMID:33773106
  • The protein has a molecular weight of approximately 52 kDa (calculated 52497 Da).

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (aa 1-200), which may target all isoforms and facilitate recognition of both active and inactive forms.
  • For Western blotting, consider lysates from human cell lines; a band at ~52 kDa is expected.
  • ELISA may be optimized using recombinant ATG4C protein as coating antigen.
  • Due to the polyclonal nature, cross-reactivity with other ATG4 family members cannot be excluded; validate in the relevant sample system.
