Biological Background: ATG4B Function and Localization

  • ATG4B, also known as Autophagin-1 or APG4B, is a cysteine protease that plays a dual role in autophagy by mediating proteolytic activation and delipidation of ATG8 family proteins. Related references: PMID:15169837, PMID:15187094, PMID:17347651

  • It is required for canonical macroautophagy, non-canonical autophagy, and mitophagy, highlighting its central role in cellular degradation pathways. Related references: PMID:33773106, PMID:33909989

  • ATG4B specifically cleaves the C-terminal amino acid of ATG8 proteins (MAP1LC3A/B/C, GABARAP, GABARAPL1/2) to expose a glycine residue necessary for conjugation to phosphatidylethanolamine (PE) and membrane insertion. Related references: PMID:15169837, PMID:15187094, PMID:17347651

  • Beyond proteolytic processing, ATG4B also acts as a deubiquitinating-like enzyme to remove ATG8 from conjugated proteins (ATG8ylation), such as ATG3, and catalyzes delipidation of PE-conjugated ATG8 proteins during autophagy. Related references: PMID:31315929, PMID:33773106, PMID:15187094, PMID:19322194, PMID:28633005

  • Among ATG4 paralogs (ATG4A-D), ATG4B is the major isoform for proteolytic activation of ATG8 proteins but exhibits weaker delipidation activity compared to others. Related references: PMID:29458288, PMID:30661429

  • In mitophagy, ATG4B can promote phagophore growth independently of its enzymatic activity and ATG8 conjugation, by regulating ATG9A trafficking and facilitating contacts between the phagophore and endoplasmic reticulum. Related references: PMID:33773106

  • ATG4B localizes predominantly to the cytoplasm, with reported associations with autophagosomes, endoplasmic reticulum, and mitochondria, consistent with its functions in autophagy and mitophagy.

  • Post-translational modifications of ATG4B include acetylation, phosphorylation, and S-nitrosylation, which may regulate its activity and stability.

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 1–393 of human ATG4B; the resulting antibody may recognize full-length ATG4B and possibly isoforms generated by alternative splicing. Validate specificity in the intended sample type.
  • For Western blotting, expect a band at approximately 44 kDa under reducing conditions. Due to potential post-translational modifications or cleavage, additional bands may appear; confirm with appropriate controls.
  • ATG4B expression is ubiquitous but may vary; consider using a positive control such as HeLa or HEK293 cell lysates, and verify knockdown if needed.
  • This product is qualified for ELISA; optimize coating concentration and detection conditions according to the assay format.
