Biological Background: ATG4A Function and Localization

  • ATG4A is a cysteine protease (autophagin-2) that plays essential roles in autophagy, including proteolytic activation and delipidation of ATG8 family proteins. Related references: PMID:12473658, PMID:15169837, PMID:17347651
  • It cleaves the C-terminal amino acid of ATG8 proteins (GABARAPL2, MAP1LC3A, GABARAP) to expose a glycine residue, a prerequisite for their conjugation to phosphatidylethanolamine and membrane insertion during autophagosome formation. Related references: PMID:12473658, PMID:15169837, PMID:17347651
  • ATG4A also functions as a deubiquitinating-like enzyme to remove ATG8 from protein conjugates (ATG8ylation), counteracting high-molecular-weight ATG8 conjugates. Related references: PMID:31315929, PMID:33773106
  • In addition to proteolytic processing, ATG4A mediates delipidation of PE-conjugated ATG8 proteins, displaying stronger delipidation activity than ATG4B. Related references: PMID:29458288, PMID:33909989
  • ATG4A contributes to phagophore growth during mitophagy independently of its protease activity, regulating ATG9A trafficking and promoting phagophore–ER contacts. Related references: PMID:33773106
  • Subcellular localization is predominantly cytoplasmic.
  • The protein is implicated in autophagy, lipid metabolism, protein transport, and the ubiquitin-like conjugation pathway, as highlighted by UniProt keywords.

Experimental Guidance and Technical Tips

  • The immunogen covers residues 289–398 of human ATG4A; consider this region when designing peptide-blocking or epitope-mapping experiments.
  • Validated for Western blot and ELISA; for immunofluorescence, immunohistochemistry, or other applications, validate in the relevant sample system with appropriate controls.
  • Given the KO validation status, this antibody may be useful for confirming target specificity in autophagy-related models; however, confirm knockout efficiency before use.
