Biological Background: ATG4A Function and Localization
- ATG4A is a cysteine protease (autophagin-2) that plays essential roles in autophagy, including proteolytic activation and delipidation of ATG8 family proteins. Related references: PMID:12473658, PMID:15169837, PMID:17347651
- It cleaves the C-terminal amino acid of ATG8 proteins (GABARAPL2, MAP1LC3A, GABARAP) to expose a glycine residue, a prerequisite for their conjugation to phosphatidylethanolamine and membrane insertion during autophagosome formation. Related references: PMID:12473658, PMID:15169837, PMID:17347651
- ATG4A also functions as a deubiquitinating-like enzyme to remove ATG8 from protein conjugates (ATG8ylation), counteracting high-molecular-weight ATG8 conjugates. Related references: PMID:31315929, PMID:33773106
