Biological Background: ATG2A Function and Localization
- Autophagy-related protein 2 homolog A (ATG2A), also known as BLTP4A, is a large lipid transfer protein essential for autophagosome formation. It tethers the isolation membrane (IM) to the endoplasmic reticulum (ER) and directly transfers phospholipids from the ER to the IM for membrane expansion. Related references: PMID:28561066, PMID:30952800, PMID:31271352
- ATG2A binds to the ER exit site (ERES) and extracts phospholipids, subsequently delivering them to ATG9 (ATG9A or ATG9B) on the expanding autophagosomal membrane. Its lipid transfer activity is strongly enhanced by WIPI1 and WDR45/WIPI4, which promote ATG2A association with phosphatidylinositol 3-monophosphate (PI3P)-rich membranes. Related references: PMID:30952800, PMID:31271352
- Beyond autophagy, ATG2A regulates the morphology and intracellular distribution of lipid droplets, linking it to lipid metabolism and storage. Related references: PMID:22219374, PMID:28561066
- Subcellularly, ATG2A localizes to the preautophagosomal structure membrane, the ER membrane, and lipid droplets, consistent with its role in membrane contact sites.
- The protein is subject to phosphorylation and alternative splicing, as indicated by its inclusion in phosphoproteomic datasets and multiple isoform predictions (UniProt Keywords: Phosphoprotein, Alternative splicing).
- With a molecular weight of approximately 213 kDa, ATG2A is a member of the ATG2 family of autophagy proteins, which are conserved mediators of membrane expansion during autophagosome biogenesis.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide derived from an internal region (aa 700–800) of human ATG2A, suggesting that the antibody may recognize both full‑length and truncated isoforms containing this sequence.
- In Western blot, a prominent band near 213 kDa is expected; however, due to the large protein size, ensure adequate transfer conditions (e.g., low‑percentage gels, extended blotting times) and use appropriate molecular weight markers.
- ELISA can be used for titer testing and semi‑quantitative analysis; consider pairing with a matching detection antibody directed against a non‑overlapping epitope.
- Cross‑reactivity is annotated only for human; reactivity with other species should be verified experimentally if needed.
- Optimal working dilutions for both WB and ELISA should be determined empirically using the relevant biological sample system (e.g., human cell lines known to express ATG2A).
