Biological Background: ATG16L1 Function and Localization
- ATG16L1 is a central autophagy hub that coordinates both canonical and non-canonical autophagy pathways through distinct functional domains. Related references: PMID:29317426 PMID:30778222
- In canonical autophagy, it forms a complex with ATG12-ATG5 to catalyze phosphatidylethanolamine (PE) conjugation to ATG8 family proteins (LC3/GABARAP), driving autophagosomal membrane elongation. Related references: PMID:23376921 PMID:23392225 PMID:24553140
- ATG16L1 also participates in non-canonical autophagy, likely catalyzing phosphatidylserine (PS) conjugation to ATG8 on single membranes, a process critical for limiting influenza A virus infection in epithelial cells. Related references: PMID:33909989
- The protein localizes to the cytoplasm, and associates with preautophagosomal structures, endosomes, and lysosomes, consistent with its roles in membrane trafficking.

- For Western blot (WB), the expected band is ~68 kDa; use appropriate positive controls and validate band identity by molecular weight markers or knockdown/knockout models.
