Biological Background: ATG12 Function and Localization

  • ATG12 (Autophagy-related protein 12) is a ubiquitin-like protein belonging to the autophagy-related (ATG) family, essential for autophagosome formation.
  • It is conjugated to ATG5 via a ubiquitin-like conjugation system involving ATG7 (E1-like) and ATG10 (E2-like), forming an ATG12-ATG5 conjugate that acts as an E3-like enzyme for ATG8 family lipidation, a critical step in autophagosome membrane expansion.
  • The ATG12-ATG5 conjugate, in complex with ATG16L1, is required for recruitment of LRRK2 to stressed lysosomes and induction of LRRK2 kinase activity under lysosomal stress conditions.
  • ATG12 is localized to the cytoplasm and preautophagosomal structure membrane.
  • Post-translational modifications include acetylation and formation of an isopeptide bond during conjugation; alternative splicing generates multiple isoforms.
  • ATG12 plays a role in host-virus interactions: it acts as a proviral factor for hepatitis C virus (HCV) by supporting translation of incoming viral RNA, and it negatively regulates the innate antiviral immune response against vesicular stomatitis virus (VSV) by impairing type I interferon production.   • The immunogen corresponds to the N-terminal region (aa 1-100) of human ATG12, which may recognize both unconjugated ATG12 (15 kDa) and the ATG12-ATG5 conjugate (~55 kDa). For Western blot, consider using reducing conditions to distinguish between these forms.
  • For IHC-P, antigen retrieval may be required; validate in tissue known to express ATG12 (ubiquitous expression, but autophagy upregulation conditions may enhance staining).
  • For IP, the antibody is suitable for capturing native ATG12 complexes; ensure proper lysis conditions to preserve protein interactions.
  • ELISA applications can be developed using this antibody as capture or detection reagent; test cross-reactivity with mouse ATG12 if needed.
