Biological Background: ATG12 Function and Localization

  • ATG12 is a ubiquitin-like protein essential for autophagy vesicle formation. It conjugates with ATG5 through a ubiquitin-like system involving ATG7 (E1-like) and ATG10 (E2-like), and the ATG12-ATG5 conjugate acts as an E3-like enzyme for ATG8 lipidation and membrane association (By similarity).
  • The ATG12-ATG5-ATG16L1 complex recruits LRRK2 to stressed lysosomes, inducing LRRK2 kinase activity upon lysosomal stress (By similarity).

  • For Western blot, validate detection around 15 kDa (or 55-60 kDa for the conjugate) in human and mouse cell lysates. Use appropriate controls for autophagy induction (e.g., starvation or lysosomal stress).
  • For ELISA, coating with the immunogen peptide or recombinant ATG12 protein may improve assay performance. Assess cross-reactivity with ATG5 and other ubiquitin-like proteins if specificity is critical.
