Biological Background: ATF6 Function and Localization
- ATF6 alpha (cyclic AMP-dependent transcription factor ATF-6 alpha) is synthesized as an endoplasmic reticulum (ER) membrane-embedded precursor that, upon ER stress, undergoes regulated intramembrane proteolysis to release the active transcription factor domain. - The processed transcription factor translocates to the nucleus and binds to ER stress response elements (ERSE: 5'-CCAAT-N9-CCAC[GA]-3' and ERSE II: 5'-ATTGG-N-CCACG-3') in cooperation with NF-Y to activate UPR target genes. - ATF6 may also participate in serum response factor-dependent transcription and is involved in foveal development and cone photoreceptor function in the retina. Related references: PMID:26029869

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 190–300 of human ATF6, a segment within the N-terminal functional domain. This design may favor detection of the transcription factor form; however, validation under ER stress conditions (e.g., tunicamycin or thapsigargin treatment) is recommended.
- In western blot (WB), the antibody may detect bands corresponding to full-length ATF6 (~90–100 kDa) or the cleaved active form (~50 kDa) depending on cellular ER stress status. Optimize sample preparation and include appropriate controls.
- For immunohistochemistry (IHC-P), consider heat-induced antigen retrieval and test the antibody at multiple dilutions in target tissue sections.
- In immunofluorescence/immunocytochemistry (IF/ICC), permeabilization is required; expect both ER and nuclear staining patterns upon UPR induction.
