Biological Background: ATF6 Function and Localization

  • ATF6, also known as activating transcription factor 6 alpha (ATF6-alpha), is a 75 kDa endoplasmic reticulum (ER) membrane-anchored precursor protein. It belongs to the ATF/CREB family of transcription factors. Related references: PMID:10564271, PMID:11158310, PMID:11779464
  • Upon ER stress, ATF6 is translocated to the Golgi where it is cleaved, releasing its N-terminal cytoplasmic domain. The processed ATF6 fragment enters the nucleus and acts as a transcription factor to activate unfolded protein response (UPR) target genes. Related references: PMID:10564271, PMID:11158310, PMID:11779464
  • ATF6 binds to ER stress response elements (ERSE) in the promoters of UPR genes, often requiring cooperation with NF-Y. It recognizes the consensus sequence 5'-CCAC[GA]-3' within ERSE motifs. Related references: PMID:10564271, PMID:11158310, PMID:11779464
  • Subcellular localization is dynamic: ATF6 is found in the ER membrane and Golgi apparatus membrane under basal conditions; its active form translocates to the nucleus upon stress induction. Related references: PMID:10564271, PMID:11158310, PMID:11779464
  • ATF6 is ubiquitously expressed and has been implicated in retinal development and cone function, with disease variants linked to achromatopsia. Related references: PMID:26029869
  • Post-translational modifications include glycosylation and ubiquitination. The protein contains a transmembrane helix and is subject to regulated intramembrane proteolysis.

Experimental Guidance and Technical Tips

  • The recombinant protein immunogen used to generate this monoclonal antibody provides high target specificity; validate in your relevant model system.
  • For Western blotting, a band of approximately 75 kDa is expected for full-length ATF6; consider using protease inhibitors to prevent degradation.
  • In ChIP assays, optimize crosslinking and sonication conditions for nuclear ATF6 binding; validate with known UPR target genes.
  • IHC-P may require antigen retrieval; test both enzymatic and heat-induced methods to ensure nuclear and/or ER staining detection.
  • Cross-reactivity with mouse allows validation in rodent models of ER stress; always include appropriate negative controls.
