Biological Background: ATF4 Function and Localization
- ATF4 is a cyclic AMP-dependent transcription factor belonging to the CREB/ATF family, also known as CREB-2, TAXREB67, and TXREB. It binds the cAMP response element (CRE) as a homodimer or heterodimer.
- Acts as the master transcription factor of the integrated stress response (ISR), induced via alternative translation re-initiation upon eIF2α phosphorylation during ER stress, amino acid starvation, or oxidative stress. Related references: PMID:31023583 PMID:32132707
- Promotes cell recovery by activating stress-responsive genes, but when associated with DDIT3/CHOP, mediates programmed cell death by inducing pro-apoptotic genes such as TRIB3 and BBC3/PUMA. Related references: PMID:15775988
- Subcellular localization includes nucleus, nuclear speckles, cytoplasm, cell membrane, and centrosome, reflecting its dynamic shuttling and diverse regulatory roles.
