Biological Background: ATF2 Function and Localization
- ATF-2 (Cyclic AMP-dependent transcription factor ATF-2), also known as CREB-2, HB16, and CRE-BP1, is a versatile transcription factor.
- It binds CRE (5'-TGACGTCA-3') and AP-1 (5'-TGACTCA-3') consensus sequences, regulating genes involved in anti-apoptosis, cell growth, and DNA damage response.
- ATF-2 shuttles between the nucleus, where it participates in global transcription and DNA repair, and the cytoplasm, where it interacts with mitochondrial outer membrane proteins HK1 and VDAC1 to promote cell death.
- The protein possesses intrinsic histone acetyltransferase (HAT) activity, acetylating histones H2B and H4 in vitro. Related references: PMID:10821277
- ATM-phosphorylated ATF-2 is critical for the DNA damage response, including ionizing radiation-induced S phase checkpoint and recruitment of the MRN complex.
- Depending on tissue and cell context, ATF-2 can exert either oncogenic or tumor suppressor functions.
- It is ubiquitously expressed, with higher abundance in the brain, and multiple isoforms are generated by alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen (aa 110-190) lies within an N-terminal region conserved across isoforms; validate isoform specificity using knockdown or knockout controls.
- For Western blotting, a band near 55 kDa is expected; include phosphatase inhibitors in lysis buffer as phosphorylation may affect migration or detection.
- In IHC-P, optimize antigen retrieval; both nuclear and cytoplasmic signals can be observed depending on the cellular state and ATF-2 localization.
- Although cross-reactivity with mouse and rat is predicted, verify performance in the relevant sample system.
- For ELISA, the recombinant immunogen can serve as a positive control to assess binding efficiency.
