Biological Background: ATF2 Function and Localization

• ATF-2 (Cyclic AMP-dependent transcription factor ATF-2), also known as CREB-2, HB16, and CRE-BP1, is a versatile transcription factor.
• It binds CRE (5'-TGACGTCA-3') and AP-1 (5'-TGACTCA-3') consensus sequences, regulating genes involved in anti-apoptosis, cell growth, and DNA damage response.
• ATF-2 shuttles between the nucleus, where it participates in global transcription and DNA repair, and the cytoplasm, where it interacts with mitochondrial outer membrane proteins HK1 and VDAC1 to promote cell death.

• In IHC-P, optimize antigen retrieval; both nuclear and cytoplasmic signals can be observed depending on the cellular state and ATF-2 localization.
• Although cross-reactivity with mouse and rat is predicted, verify performance in the relevant sample system.
