Biological Background: ATAD3A/B Function and Localization

  • ATAD3A is an ATPase family AAA domain-containing protein, existing in multiple isoforms via alternative splicing.
  • Localizes to the mitochondrial inner membrane, matrix, and nucleoids, playing a critical role in mitochondrial network organization, metabolism, and cell growth.
    Related references: PMID:17210950, PMID:20154147, PMID:22453275
  • May participate in mitochondrial protein synthesis and DNA replication, likely by binding to mitochondrial DNA D-loops and stabilizing nucleoids.
    Related references: PMID:22453275, PMID:17210950
  • Required for enhanced cholesterol channeling during hormone-dependent steroidogenesis.
    Related references: PMID:22453275
  • Involved in mitochondrial-mediated antiviral innate immunity.
    Related references: PMID:31522117
  • Protects mitochondria from PERK-mediated unfolded protein response by inhibiting EIF2AK3/PERK at ER-mitochondria contact sites; this function is independent of its ATPase activity.
    Related references: PMID:39116259
  • Participates in mitochondrial DNA damage response, promoting signaling between damaged genomes and the membrane to activate the integrated stress response (ISR).
    Related references: PMID:37832546
  • Overexpressed in lung adenocarcinomas at the protein level, suggesting a role in tumorigenesis.

Experimental Guidance and Technical Tips

  • The immunogen used is a recombinant fusion protein covering amino acids 1–240 of human ATAD3A/B, a conserved N-terminal region; this design may support cross-reactivity with both ATAD3A and ATAD3B, as well as with mouse and rat orthologs.
  • For Western blot, consider testing multiple tissue lysates (human, mouse, rat) and note the predicted molecular weights of 57, 66, and 71 kDa, which correspond to ATAD3A/B isoforms; validate specificity with appropriate controls.
  • In IHC-P, optimize antigen retrieval conditions and validate in tissues known to express ATAD3A, such as lung adenocarcinoma, where overexpression is reported.
  • For ELISA, use as a capture or detection antibody in combination with a suitable paired reagent; verify performance under standard assay conditions.
