Biological Background: ATAD3A/ATAD3B Function and Localization
- ATAD3A (ATPase family AAA domain-containing protein 3A) and its paralog ATAD3B are mitochondrial membrane proteins with essential roles in mitochondrial dynamics and cell metabolism.
- ATAD3A is essential for mitochondrial network organization, metabolism, and cell growth. Related references: PMID:17210950, PMID:20154147, PMID:22453275
- It participates in mitochondrial protein synthesis and may contribute to mitochondrial DNA replication and nucleoid stability. Related references: PMID:17210950, PMID:22453275
- ATAD3A is required for cholesterol trafficking to facilitate hormone-dependent steroidogenesis. Related references: PMID:22453275
- It plays a role in mitochondrial-mediated antiviral innate immunity. Related references: PMID:31522117
- ATAD3A protects mitochondria from ER stress by inhibiting PERK at contact sites, independent of its ATPase activity. Related references: PMID:39116259
- It is involved in the mitochondrial DNA damage response, promoting activation of the integrated stress response. Related references: PMID:37832546
- ATAD3A localizes to the mitochondrial inner membrane, matrix, and nucleoid; it is overexpressed in lung adenocarcinomas and subject to acetylation and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1–240) of human ATAD3B, which shares high sequence similarity with ATAD3A, enabling detection of both proteins. Validate specificity in your target sample system.
- For Western blot, consider using mitochondrial-enriched extracts from human, mouse, or rat tissues or cells; the predicted molecular weights are 71 kDa (ATAD3A) and 73 kDa (ATAD3B).
- In immunofluorescence/ICC, co-stain with a mitochondrial marker to confirm subcellular localization.
- For ELISA applications, pair with a capture antibody if developing a sandwich assay, and optimize blocking and detection conditions.
