Biological Background: ASL Function and Localization
- ASL encodes argininosuccinate lyase, a homotetrameric enzyme of the urea cycle that catalyzes the reversible cleavage of L-argininosuccinate to fumarate and L-arginine, a critical step for hepatic nitrogen detoxification and de novo arginine synthesis in nonhepatic tissues. - ASL is an essential regulator of intracellular and extracellular L-arginine pools.
- In the citrulline-nitric oxide cycle, ASL assembles into tissue-specific multiprotein complexes with argininosuccinate synthase (ASS1), the cationic amino acid transporter SLC7A1, and nitric oxide synthase isoforms (NOS1, NOS2, or NOS3) to enable cell-autonomous L-arginine synthesis and channel extracellular L-arginine into nitric oxide production. - Alternative splicing generates multiple ASL transcript variants, and the protein undergoes post-translational modifications such as acetylation.

Experimental Guidance and Technical Tips
- For Western blot (WB), the expected band for endogenous ASL is around 52 kDa; consider optimizing primary antibody dilution and using appropriate loading controls.
- For immunohistochemistry paraffin-embedded tissues (IHC-P), heat-induced antigen retrieval with citrate buffer (pH 6.0) is recommended to unmask epitopes.
- In immunofluorescence/immunocytochemistry (IF/ICC), fix cells with 4% paraformaldehyde and permeabilize with 0.1% Triton X-100 before staining; counterstaining with nuclear dyes may help localize the cytosolic enzyme.
- As a polyclonal antibody, it may recognize multiple epitopes and potential isoforms; validate specificity in the relevant species and tissue system.
- For ELISA, the antibody can be used for antigen capture or detection; appropriate coating and blocking conditions should be determined empirically.
