Biological Background: ASGR1 Function and Localization
- ASGR1 (Asialoglycoprotein receptor 1), also known as hepatic lectin H1 (HL-1) and CLEC4H1, belongs to the C-type lectin family. It is a transmembrane protein predominantly expressed on hepatocytes.
- Functions as a key endocytic receptor: specifically binds terminal galactose and N-acetylgalactosamine residues on desialylated glycoproteins, mediating their clearance from circulation.
- Plays a critical role in the removal of desialylated platelets via a feedback loop that regulates thrombopoietin (TPO) expression through JAK2-STAT3 signaling. Related references: PMID:25485912
- Contributes to the recognition and elimination of activated lymphocytes, with hepatocytes acting as cytotoxic effectors in this process. Related references: PMID:21656538

- The immunogen corresponds to a recombinant protein spanning amino acids 62–291 of human ASGR1 (NP_001662.1). This region covers part of the extracellular domain involved in carbohydrate recognition, making the antibody suitable for detecting both membrane-bound and secreted forms.
- For Western blotting (WB), a band around 33 kDa is expected. Consider using reducing conditions; validate with positive control lysates from liver tissue or HepG2 cells.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF-P), paraffin-embedded or frozen liver sections are appropriate; antigen retrieval may be necessary.
- ELISA application is supported; optimal dilution should be determined empirically.
