Biological Background: ASGR1 Function and Localization
- ASGR1 encodes the asialoglycoprotein receptor 1 (ASGPR1), also known as C-type lectin domain family 4 member H1 (CLEC4H1) or hepatic lectin H1 (HL-1), a transmembrane protein predominantly expressed on hepatocytes.
- Mediates the endocytosis of desialylated plasma glycoproteins, contributing to the clearance of aged platelets and activated lymphocytes, and maintaining serum glycoprotein homeostasis. Related references: PMID:25485912
- Functions in platelet homeostasis by activating a JAK2-STAT3 feedback loop that regulates thrombopoietin (TPO) expression upon binding desialylated platelets.
- Participates in immune surveillance by recognizing and eliminating activated lymphocytes; hepatocytes act as cytotoxic effectors during this process. Related references: PMID:21656538
- May also play a role in lipid homeostasis (by similarity).
- Acts as an alternate receptor for ACE2-independent SARS-CoV-2 entry, highlighting its role in host-virus interactions.
- Subcellular localization: primarily found on the cell membrane; also detected as a secreted form.
- Tissue specificity: expression is restricted to hepatic parenchymal cells, making it a liver-specific marker.
- Key structural features include a C-type lectin domain, calcium-dependent activity, a transmembrane helix, and post-translational modifications such as palmitoylation, phosphorylation, and disulfide bond formation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1–150 of human ASGR1 (NP_001662.1), covering the N-terminal extracellular region, which may enable detection of both membrane-bound and secreted forms.
- For Western blotting (WB), the predicted molecular weight is approximately 33 kDa; however, glycosylation may result in a higher observed band. Validate in the relevant sample system.
- For immunohistochemistry (IHC-P), strong signal is expected in liver tissue sections. Include appropriate positive (liver) and negative (non-hepatic) tissue controls.
- For ELISA, consider pairing this antibody with another clone or a monoclonal antibody for sandwich assay development; perform cross-reactivity testing.
- Cross-reactivity with mouse and rat has been confirmed; for other species, validation is recommended before use.
