Biological Background: ASC/TMS1 Function and Localization

  • Apoptosis-associated speck-like protein containing a CARD (ASC/TMS1) is a key mediator of apoptosis and inflammation. It promotes caspase-mediated apoptosis involving predominantly caspase-8 and also caspase-9 in a probable cell type-specific manner. Related references: PMID:11103777, PMID:12646168
  • ASC functions as an integral adapter in the assembly of various inflammasomes (NLRP1, NLRP2, NLRP3, NLRP6, AIM2 and probably IFI16), which recruit and activate caspase-1 leading to processing and secretion of pro-inflammatory cytokines.   • Through its pyrin and CARD domains, ASC mediates homotypic interactions that nucleate the formation of caspase-1 filaments, acting as a platform for caspase-1 polymerization and macrophage pyroptosis. Related references: PMID:14499617, PMID:19234215, PMID:24630722
  • ASC is localized to the cytoplasm, nucleus, endoplasmic reticulum, mitochondria, and Golgi apparatus membrane; it is recruited to inflammasomes upon activation.

Experimental Guidance and Technical Tips

  • The immunogen covers the N-terminal 91 amino acids, which includes the pyrin domain (PYD); the antibody may recognize both full-length and any N-terminal isoforms. For applications requiring detection of specific isoforms, validation with appropriate controls is recommended.
  • For Western blot (WB), a primary band around 22 kDa is expected; however, potential post-translational modifications or cleavage fragments may produce additional bands; loading controls and molecular weight markers should be used.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), permeabilization and fixation conditions should be optimized, as ASC localizes to multiple subcellular compartments. Nuclear and cytoplasmic staining patterns may vary depending on activation state.
  • Immunoprecipitation (IP) and ELISA applications are also validated; consider using fresh lysates and including protease inhibitors to preserve protein interactions.
  • Cross-reactivity with mouse and rat is reported; however, sequence homology should be checked for studies in these species, and pilot experiments are advised.
