Biological Background: ASC/TMS1 Function and Localization

  • ASC/TMS1, also known as PYCARD, CARD5, or TMS1, is a 22 kDa protein containing pyrin (PYD) and caspase recruitment (CARD) domains, critical for apoptosis and inflammation. Related references: PMID:11103777 PMID:12646168
  • It functions as an essential adaptor in various inflammasomes (NLRP1, NLRP3, AIM2, etc.), recruiting caspase-1 to promote maturation of IL-1β and IL-18 and pyroptotic cell death. Related references: PMID:15030775 PMID:16982856 PMID:24630722

  • The protein is widely expressed at low levels in many tissues including blood leukocytes, lung, small intestine, and at lower levels in placenta, liver, kidney; very low in skeletal muscle, heart, brain.
