Biological Background: ASB4 Function and Localization

  • ASB4 (Ankyrin repeat and SOCS box protein 4) is a member of the SOCS box protein family, containing ankyrin repeats and a SOCS box domain. It functions as a substrate-recognition component of a SCF-like ECS E3 ubiquitin-protein ligase complex, mediating ubiquitination and proteasomal degradation of target proteins.
  • ASB4 negatively regulates insulin receptor substrate 4 (IRS4) by directing its ubiquitination and degradation, thereby attenuating downstream IRS4 signaling. Related references: PMID:21955513
  • In vascular development, ASB4 plays a critical role in early vasculogenesis and placentation by targeting the transcriptional regulator inhibitor of DNA binding 2 (ID2) for polyubiquitination and proteasome-dependent degradation. Related references: PMID:24586788
