Biological Background: ASB4 Function and Localization
- ASB4 (Ankyrin repeat and SOCS box protein 4) is a member of the SOCS box protein family, containing ankyrin repeats and a SOCS box domain. It functions as a substrate-recognition component of a SCF-like ECS E3 ubiquitin-protein ligase complex, mediating ubiquitination and proteasomal degradation of target proteins.
- ASB4 negatively regulates insulin receptor substrate 4 (IRS4) by directing its ubiquitination and degradation, thereby attenuating downstream IRS4 signaling. Related references: PMID:21955513
- In vascular development, ASB4 plays a critical role in early vasculogenesis and placentation by targeting the transcriptional regulator inhibitor of DNA binding 2 (ID2) for polyubiquitination and proteasome-dependent degradation. Related references: PMID:24586788
- ASB4 also acts as a negative regulator of GPS1, a component of the COP9 signalosome (CSN) multiprotein complex, thereby inhibiting the serine phosphorylation of IRS1 (By similarity).
- Subcellular localization: ASB4 is predominantly cytoplasmic.
- Keywords indicate alternative splicing, hydroxylation, and involvement in the ubiquitin conjugation pathway.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-300) of human ASB4. Cross-reactivity with mouse is reported; for other species, verification is recommended.
- This antibody is suitable for IHC-P and ELISA. Optimal working dilutions should be determined empirically for each assay.
- Consider using appropriate controls (e.g., ASB4 knockdown or knockout lysates) to confirm specificity in your experimental system.
