Biological Background: ASB2 Function and Localization
- ASB2 functions as a substrate-recognition component of an SCF-like ECS (Elongin-Cullin-SOCS-box protein) E3 ubiquitin-protein ligase complex, mediating ubiquitination and subsequent proteasomal degradation of target proteins. (PubMed:15590664, PubMed:16325183) Related references: PMID:15590664 PMID:16325183
- It participates in Notch-induced signaling by promoting ubiquitination and degradation of substrates such as TCF3/E2A and JAK2 (PubMed:21119685). During embryonic development, ASB2 is crucial for complete heart looping and cardiomyocyte differentiation (PubMed:32179481). Related references: PMID:21119685 PMID:32179481
- ASB2 specifically targets SMAD9 for proteasomal degradation, regulating its cellular abundance to prevent excessive accumulation (PubMed:34845242). Related references: PMID:34845242
- In myogenic differentiation, ASB2 targets filamin FLNB and the intermediate filament protein DES for degradation (PubMed:19300455), acting as a negative regulator of skeletal muscle mass. Related references: PMID:19300455
- ASB2 mediates the degradation of filamins FLNA and FLNB (PubMed:21737450; PubMed:22916308; PubMed:24044920), enhancing hematopoietic cell adhesion to fibronectin (PubMed:21737450) and facilitating dendritic cell migration. In hematopoietic cells, ASB2 does not promote degradation of JAK1 or JAK2 (PubMed:22916308). Related references: PMID:21737450 PMID:22916308 PMID:24044920
- Subcellularly, ASB2 localizes to the cytoplasm, associating with cytoskeletal structures such as stress fibers and the sarcomeric Z-line in myofibrils.
- ASB2 is predominantly expressed in muscle cells and hematopoietic cells.
- The protein contains ankyrin repeat domains and a SOCS box, characteristic of the ASB family, and undergoes alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (amino acids 448–587) of human ASB2, which may provide specificity for full-length ASB2. Cross-reactivity with rat suggests conserved epitopes.
- This polyclonal antibody is applicable for Western blotting (WB) and ELISA. For WB, consider using whole-cell lysates from human or rat muscle/hematopoietic cell lines, and validate band specificity around the predicted molecular weight of 70 kDa.
- Optimize primary antibody dilutions and incubation conditions empirically for your specific assay and sample type. Include appropriate positive and negative controls.
- If using for ELISA, assess coating antigen and detection conditions to minimize background and maximize signal-to-noise ratio.
