Biological Background: ART1 Function and Localization
- ART1 (ADP-ribosyltransferase 1) is a GPI-linked NAD(P)+-arginine ADP-ribosyltransferase, also designated ARTC1 or CD296.
- It belongs to the ADP-ribosyltransferase C2/C3 toxin-like family of mono(ADP-ribosyl)transferases that utilize NAD+ or NADP+ as substrates.
- ART1 catalyzes the transfer of ADP-ribose from NAD+ to arginine residues on target proteins; a known substrate is the glucagon-like peptide 1 receptor (GLP1R).
- The enzyme is anchored to the outer leaflet of the sarcoplasmic reticulum membrane via a C-terminal glycosylphosphatidylinositol (GPI) moiety.
- The primary translation product contains a signal peptide and undergoes N-linked glycosylation, disulfide bond formation, and GPI-attachment during secretion pathway processing.
- Key functional and structural features include its glycoprotein nature, GPI-anchor, lipid modification, and dependence on NAD/NADP cofactors.
Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 23–295 of human ART1, covering the N-terminal catalytic domain; this design supports high specificity for the mature protein.
- For western blotting, a band at approximately 36 kDa is expected (calculated molecular weight 36.3 kDa); validate detection using relevant human and mouse cell lines or tissues.
- In immunohistochemistry and immunofluorescence, optimize fixation and permeabilization protocols due to the membrane-associated GPI-anchored localization.
- The polyclonal nature may yield robust signal amplification; however, lot-to-lot consistency should be verified for quantitative applications.
- Detection across human and mouse species is documented; cross-reactivity with other organisms should be tested empirically if needed.
