Biological Background: ARPC2 Function and Localization
- ARPC2 (Actin-related protein 2/3 complex subunit 2), also known as p34-ARC, is a 34 kDa subunit of the Arp2/3 complex, a key regulator of actin dynamics.
- The Arp2/3 complex mediates actin polymerization upon stimulation by nucleation-promoting factors (NPFs), generating branched actin networks that provide force for cell motility.

PMID:9230079 - Beyond cytoskeletal functions, ARPC2 and the Arp2/3 complex promote actin polymerization in the nucleus, where they regulate gene transcription and DNA damage repair.

- ARPC2 is subject to acetylation, and its three-dimensional structure has been determined, providing insights into its function.
Experimental Guidance and Technical Tips
- The immunogen used is a synthetic peptide spanning amino acids 200–300 of human ARPC2. This region is predicted to be surface-exposed and specific to ARPC2, which may contribute to antibody specificity.
- For Western blotting, the antibody detects a band consistent with the predicted molecular weight of ~34 kDa. Use appropriate controls to confirm target identity.
- In immunofluorescence applications, ARPC2 is expected to localize to both the cytoplasm and nucleus, with enrichment at cell protrusions and synaptic structures.
- For immunoprecipitation, this antibody can be used to pull down the Arp2/3 complex; co-immunoprecipitation of other complex subunits (e.g., ARPC1, ARPC3) can serve as a validation.
