Biological Background: Arp2 Function and Localization
- Arp2 (Actin-related protein 2), also known as Actin-like protein 2, is encoded by the ACTR2 gene.
- Arp2 is an ATP-binding component of the Arp2/3 complex, which mediates actin polymerization upon stimulation by nucleation-promoting factors, driving the formation of branched actin networks in the cytoplasm for cell motility. Related references: PMID:9000076
- In podocytes, Arp2 is required for lamellipodia formation downstream of AVIL and PLCE1 regulation. Related references: PMID:29058690
- The Arp2/3 complex also promotes actin polymerization in the nucleus, regulating gene transcription and repair of damaged DNA. Related references: PMID:17220302, PMID:29925947
- Nuclear Arp2/3 complex promotes homologous recombination repair by driving motility of double-strand breaks. Related references: PMID:29925947
- Arp2 localizes to the cytoplasm, cytoskeleton, cell projections, and nucleus.
- Post-translational modifications include acetylation, and alternative splicing generates multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen comprises the N-terminal region (aa 1-160) of human Arp2, which may help recognize both native and denatured protein conformations.
- For Western blotting, expect a band near 45 kDa; optimize loading controls and detection conditions.
- In immunohistochemistry and immunofluorescence, validate specificity using appropriate tissue/cell controls and consider signal localization patterns.
- Given reported cross-reactivity with mouse and rat, the antibody is suitable for comparative studies, but always confirm in the relevant model system.
