Biological Background: Aromatase (CYP19A1) Function and Localization

  • Aromatase (CYP19A1) is a cytochrome P450 monooxygenase that converts C19 androgens (androstenedione and testosterone) to C18 estrogens (estrone and estradiol) through three successive oxidations. Related references: PMID:27702664 PMID:2848247
  • The enzyme catalyzes three successive oxidations: two conventional oxidations at C19 to 19-hydroxy and 19-oxo/19-aldehyde intermediates, then a third aromatization step resulting in phenolic A ring and formic acid, or alternatively producing a 19-norsteroid. Related references: PMID:20385561
  • CYP19A1 also converts dihydrotestosterone to delta1,10-dehydro 19-nordihydrotestosterone, potentially regulating androgen homeostasis, and displays 2-hydroxylase activity toward estrone. Related references: PMID:22773874
  • Subcellularly, aromatase localizes to the endoplasmic reticulum membrane and microsome membrane.
  • The protein is widely expressed in tissues, including adult and fetal brain, placenta, skin fibroblasts, adipose tissue, and gonads.
  • Keywords: alternative splicing, disease variant, heme and iron binding, lipid metabolism, transmembrane helix. Mutations in CYP19A1 are associated with pseudohermaphroditism.

Experimental Guidance and Technical Tips

  • This rabbit monoclonal antibody was raised against a synthetic peptide corresponding to the N-terminal region (amino acids 1-100) of human aromatase, which may provide specificity for intact CYP19A1.
  • For Western blotting, a band at approximately 58 kDa is expected; consider using appropriate positive controls (e.g., placental tissue lysate) and validate the molecular weight in your system.
  • Immunofluorescence and immunohistochemistry may reveal endoplasmic reticulum-like staining; co-localization with ER markers is recommended to confirm specificity.
  • The antibody's reactivity with mouse and rat orthologs is listed; if using these species, verify sequence homology in the epitope region to ensure cross-reactivity.
  • For ELISA applications, pair this antibody with a matched detection antibody or use it as a capture/detection reagent, and validate with recombinant aromatase or tissue extracts.
