Biological Background: MANF Function and Localization
- MANF (mesencephalic astrocyte-derived neurotrophic factor), also known as ARMET (arginine-rich protein) or ARP, is a secreted protein that resides in the endoplasmic reticulum (ER) and sarcoplasmic reticulum lumen.
- Selectively promotes the survival of dopaminergic neurons of the ventral mid-brain, playing a critical role in neuroprotection. Related references: PMID:12794311
- Modulates GABAergic transmission to dopaminergic neurons of the substantia nigra, enhancing both spontaneous and evoked inhibitory postsynaptic currents.
- Inhibits cell proliferation and protects against endoplasmic reticulum (ER) stress-induced cell death by stabilizing the ER chaperone HSPA5/BiP and suppressing the unfolded protein response (UPR). Related references: PMID:18561914 PMID:22637475 PMID:29497057
- Upregulated and secreted in response to ER stress and hypoxia; mediates cellular uptake via binding to the lipid sulfatide (3-O-sulfogalactosylceramide) on target cell membranes, which is required for alleviating ER stress and cytotoxicity. Related references: PMID:22637475 PMID:29497057
- Essential for postnatal development processes including lung maturation, splenic white pulp architecture, skeletal muscle repair, myocardial protection, and endochondral ossification.
- Key structural features include a signal peptide, disulfide bonds, glycosylation, and phosphorylation sites, with a mature molecular weight of approximately 20.7 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 25–182 of human MANF, covering the mature protein after signal peptide cleavage, which is conserved across human, mouse, and rat.
- Suitable for western blot (WB) under reducing conditions, where a band around 21 kDa is expected; consider optimizing antibody dilution and exposure time based on sample expression levels.
- For ELISA, both direct and sandwich formats may be explored; antigen coating conditions and detection antibody concentration should be validated in the relevant sample system.
- Cross-reactivity with mouse and rat allows for multispecies experimental designs; always include appropriate species-matched controls.
