Biological Background: ARL6 Function and Localization

  • ARL6 (ADP-ribosylation factor-like protein 6, also known as BBS3) is a small GTP-binding protein belonging to the ARF family.
  • It functions in membrane protein trafficking at the ciliary base by recruiting the BBSome coat complex to the plasma membrane, a process essential for sorting proteins to primary cilia. Related references: PMID:20603001
  • Together with BBS1 and the BBSome complex, ARL6 regulates trafficking of PKD1 (by similarity) and SMO to cilia, thereby modulating sonic hedgehog (SHH) signaling. Related references: PMID:22072986
  • ARL6 is implicated in ciliary assembly/disassembly and Wnt signaling cascades. Related references: PMID:20207729
  • It localizes to the cilium membrane, basal body, and axoneme, and is also present in the cytoplasm.
  • Myristoylated and GTP-binding, ARL6 requires magnesium for its activity. Alternative splicing generates an isoform required for proper retinal function (by similarity). Mutations in ARL6 cause Bardet-Biedl syndrome 3, characterized by ciliopathy-related phenotypes including retinal degeneration, obesity, and intellectual disability.
