Biological Background: ARL3 Function and Localization
- ARL3 is a small GTP-binding protein that cycles between inactive GDP-bound and active GTP-bound forms, with the rate of cycling regulated by guanine nucleotide exchange factors (GEF) and GTPase-activating proteins (GAP). Related references: PMID:16525022 PMID:18588884
- It is required for normal cytokinesis and cilia signaling, and for targeting proteins to the cilium, including myristoylated NPHP3 and prenylated INPP5E. Related references: PMID:22085962 PMID:30269812

- In IF/ICC, ARL3 is expected to localize to the cilium and centrosome; co-staining with ciliary markers (e.g., acetylated tubulin) is advisable.
- For ELISA, the antibody can be used to detect recombinant or native ARL3; assay conditions should be optimized.
