Biological Background: ARIH1 Function and Localization
- ARIH1 (also known as HHARI, ARI-1, UbcH7-binding protein, MOP-6) is an E3 ubiquitin-protein ligase.
- It catalyzes ubiquitination of target proteins together with ubiquitin-conjugating enzyme E2 UBE2L3. Related references: PMID:15236971, PMID:21532592, PMID:23707686
- Acts as an atypical E3 ligase by associating with cullin-RING ubiquitin ligase (CRL) complexes, mediating the initial ubiquitin on CRL substrates, which is elongated by CDC34/UBE2R1 and UBE2R2. Related references: PMID:27565346
- Ligase activity is activated upon binding to neddylated cullin-RING ubiquitin ligase complexes. Related references: PMID:24076655, PMID:27565346
- Plays a role in protein translation in response to DNA damage by mediating ubiquitination of EIF4E2; reported outcomes include promotion of cap-binding and translation arrest or degradation. Also mediates ISGylation of EIF4E2. Related references: PMID:25624349, PMID:14623119, PMID:17289916
- In vitro, controls degradation of SUN2, a component of the LINC complex, potentially influencing nuclear morphology and localization. Related references: PMID:29689197
- Contains zinc-finger domains and is involved in the ubiquitin conjugation pathway as a metal-binding transferase.
- Widely expressed across human tissues, with localization to the cytoplasm, nucleus, and Cajal bodies.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning the C-terminal region (amino acids 400–557) of human ARIH1. This region shows high sequence homology across human, mouse, and rat, consistent with the reported cross-reactivity.
- For Western blotting, ARIH1 has a predicted molecular weight of ~64 kDa. Use standard whole-cell or tissue lysates; validate the band size in your system, as post-translational modifications may cause shifts.
- As a polyclonal antibody, ensure consistent lot-to-lot performance by including appropriate positive and negative controls in each experiment.
- For ELISA, consider direct antigen coating or sandwich formats, optimizing antibody dilution and secondary detection conditions.
