Biological Background: ARID1B Function and Localization
- AT-rich interactive domain-containing protein 1B (ARID1B), also designated BAF250B, hOsa2, or p250R, is a subunit of the SWI/SNF ATP-dependent chromatin remodeling complex.
- Core function: ARID1B participates in transcriptional activation and repression by altering DNA–histone contacts within nucleosomes, thereby modulating chromatin accessibility.
- Neural development role: ARID1B is a component of both neural progenitors-specific (npBAF) and neuron-specific (nBAF) complexes, essential for the self-renewal of neural stem cells and later for the regulation of dendrite growth genes.
- ARID1B binds DNA non-specifically, contributing to the recruitment or stabilization of SWI/SNF complexes on chromatin. Related references: PMID:14982958, PMID:15170388
- Subcellular localization: ARID1B is predominantly nuclear, consistent with its chromatin-associated functions.
- Tissue expression: Widely expressed, with highest levels observed in heart, skeletal muscle, and kidney.
- Post-translational modifications and keywords: The protein undergoes acetylation, methylation, and phosphorylation, and is also subject to alternative initiation and splicing.
- Disease association: ARID1B mutations are linked to Coffin-Siris syndrome (CSS1) and autosomal dominant intellectual disability (MRD12), underscoring its critical role in neurodevelopment.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a central region (amino acids 400–650), which may help minimize cross-reactivity with homologous family members. Verify sequence alignment for your species of interest.
- For Western blotting, a high molecular weight band around 244 kDa is expected. Use low-percentage polyacrylamide gels (e.g., 6–8%) and optimize transfer conditions for large proteins.
- In IHC-P and IF/ICC, nuclear staining is anticipated. Include appropriate isotype controls and validate in relevant human, mouse, or rat tissues.
- For ELISA applications, the polyclonal nature may provide robust signal. Perform bridging studies to ensure lot-to-lot consistency if used in quantitative assays.
