Biological Background: ARID1A Function and Localization
- ARID1A (AT-rich interactive domain-containing protein 1A), also known as BAF250a, is a key subunit of the SWI/SNF chromatin remodeling complex. It plays a critical role in altering DNA-histone contacts within nucleosomes in an ATP-dependent manner, thereby regulating transcription.
- The protein contains an ARID domain that binds DNA non-specifically, facilitating the targeting of SWI/SNF complexes to chromatin. It is involved in both transcriptional activation and repression of select genes.
- ARID1A is a component of neural progenitor-specific (npBAF) and neuron-specific (nBAF) chromatin remodeling complexes. During neural development, the switch from npBAF to nBAF is essential for proper neuronal differentiation and dendrite growth.
- Localized strictly to the nucleus, ARID1A exhibits tissue-specific expression, with high levels in spleen, thymus, prostate, testis, ovary, small intestine, colon, and peripheral blood leukocytes, and lower expression in other tissues.
- Post-translational modifications of ARID1A include acetylation and phosphorylation, which may regulate its activity or interactions within the SWI/SNF complex.
- Alternative splicing of ARID1A produces multiple isoforms, potentially expanding the functional diversity of SWI/SNF complexes.
Experimental Guidance and Technical Tips
- This monoclonal antibody was raised against a recombinant fusion protein corresponding to amino acids 1204-1314 of human ARID1A. It recognizes a region within the C-terminal portion, which may be present in certain isoforms. Verify the target isoforms in your experimental system.
- For immunohistochemistry (IHC-P), optimize antigen retrieval conditions and antibody dilution. Use appropriate positive and negative tissue controls, as expression levels vary across tissues.
- In immunofluorescence (IF/ICC), fix and permeabilize cells appropriately. Nuclear counterstaining is recommended to confirm the expected nuclear localization.
- For ELISA, ensure proper coating conditions for the recombinant immunogen or target protein. The antibody may be used as a capture or detection reagent in sandwich ELISA formats.
