Biological Background: ARHGEF5 Function and Localization

  • ARHGEF5 functions as a guanine nucleotide exchange factor (GEF) that strongly activates RHOA and RHOB, weakly activates RHOC and RHOG, and shows no effect on RHOD, RHOV, RHOQ, or RAC1. Through this activity, it regulates cell shape and actin cytoskeletal organization. (PubMed:15601624) Related references: PMID:15601624
  • The protein plays a role in actin dynamics by promoting the loss of actin stress fibers and the formation of membrane ruffles and filopodia. (PubMed:14662653) Related references: PMID:14662653
  • ARHGEF5 is classified as a proto-oncogene and contains an SH3 domain, indicating potential roles in signal transduction and oncogenesis.
  • The protein localizes to the cytoplasm, nucleus, and cell projections such as podosomes, implicating it in cell adhesion and migration structures.
  • It is ubiquitously expressed, with highest levels in placenta, and significant expression in colon, kidney, trachea, prostate, liver, pancreas, pituitary, thyroid, and mammary glands. In fetal tissues, high expression is seen in kidney, lung, and liver.
  • Alternative splicing produces multiple isoforms, and the protein undergoes post-translational modifications including phosphorylation and methylation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 1248–1597 of human ARHGEF5, a C-terminal region. For IHC-P, consider appropriate antigen retrieval conditions and validate in positive tissues such as placenta or colon, where high expression is reported.
  • For ELISA, standard direct or indirect protocols may be employed. Optimize coating antigen concentration and use appropriate blocking buffers to minimize background.
  • This polyclonal antibody recognizes human, mouse, and rat ARHGEF5. Cross-reactivity with other Rho GEF family members cannot be excluded; verify specificity using knockdown or knockout controls where possible.
  • As ARHGEF5 is involved in podosome formation and cell migration, functional validation in relevant cell models (e.g., dendritic cells, SRC-transformed cells) may be beneficial.
