Biological Background: ARHGEF5 Function and Localization
- ARHGEF5 functions as a guanine nucleotide exchange factor (GEF) that strongly activates RHOA and RHOB, weakly activates RHOC and RHOG, and shows no effect on RHOD, RHOV, RHOQ, or RAC1. - ARHGEF5 is classified as a proto-oncogene and contains an SH3 domain, indicating potential roles in signal transduction and oncogenesis.
- The protein localizes to the cytoplasm, nucleus, and cell projections such as podosomes, implicating it in cell adhesion and migration structures.
- It is ubiquitously expressed, with highest levels in placenta, and significant expression in colon, kidney, trachea, prostate, liver, pancreas, pituitary, thyroid, and mammary glands. In fetal tissues, high expression is seen in kidney, lung, and liver.

- For ELISA, standard direct or indirect protocols may be employed. Optimize coating antigen concentration and use appropriate blocking buffers to minimize background.
- This polyclonal antibody recognizes human, mouse, and rat ARHGEF5. Cross-reactivity with other Rho GEF family members cannot be excluded; verify specificity using knockdown or knockout controls where possible.
