Biological Background: ARHGEF3 Function and Localization

  • ARHGEF3 (Rho guanine nucleotide exchange factor 3) is also known as Exchange factor found in platelets and leukemic and neuronal tissues (XPLN), and is a member of the Dbl family of GEFs.
  • It functions as a guanine nucleotide exchange factor (GEF) specifically activating the small GTPases RhoA and RhoB, thereby promoting their transition from an inactive GDP-bound state to an active GTP-bound state.
  • By activating RhoA and RhoB, ARHGEF3 regulates actin cytoskeleton dynamics, cell morphology, migration, and various signaling pathways.
  • The protein is widely expressed across human tissues, with highest levels observed in adult brain and skeletal muscle, and lower levels in heart and kidney.
  • ARHGEF3 localizes predominantly to the cytoplasm.
  • Post-translational modifications include phosphorylation, and the gene undergoes alternative splicing.
  • It is identified in proteomic studies and is a reference proteome member.

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 287-526 of ARHGEF3, which encompasses a region likely critical for GEF activity; this may provide specific detection of the target.
  • For Western blot, consider using whole cell lysates from human or mouse tissues/cell lines expressing ARHGEF3 (e.g., brain, skeletal muscle) and validate band size around 60 kDa.
  • This polyclonal antibody may also be suitable for ELISA; optimize coating and detection conditions based on your assay requirements.
  • Cross-reactivity with mouse has been confirmed, so consider appropriate sample species during experimental design.
