Biological Background: ARHGAP11B Function and Localization
- ARHGAP11B is an inactive Rho GTPase-activating protein 11B, also referred to as Rho-type GTPase-activating protein 11B.
- This hominin-specific protein is essential for evolutionary expansion of the brain neocortex, promoting basal progenitor amplification and increasing neuron production during fetal corticogenesis. Related references: PMID:25721503 PMID:27957544 PMID:30484771
- It promotes basal progenitor proliferation by inhibiting the mitochondrial permeability transition pore (mPTP) through interaction with ADP:ATP translocase, thereby raising mitochondrial Ca²⁺ levels and driving glutamine catabolism. Related references: PMID:31883789
- The lack of GTPase activator activity is not a defect but a prerequisite for its role in basal progenitor amplification. Related references: PMID:25721503 PMID:27957544
- ARHGAP11B localizes to the mitochondrial matrix, consistent with its involvement in mPTP regulation.
- Annotated keywords include GTPase activation, Mitochondrion, Reference proteome, and Transit peptide.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 218–267, a region unique to human ARHGAP11B; ensure sequence alignment if using non-human samples.
- For WB, consider using whole cell lysates from human brain tissue or neuron-derived cell lines, where target expression is expected.
- Given the hominin specificity, cross-reactivity with rodent or other model organisms is unlikely; always include species-appropriate controls to verify reactivity.
- As a polyclonal antibody, batch-to-batch consistency should be validated for critical quantitative applications.
- Optimize antibody dilution and incubation conditions in your specific experimental system; recommended starting dilutions are not provided and must be determined empirically.
