Biological Background: Argonaute-2 Function and Localization
- Protein Argonaute-2 (AGO2, also known as eIF2C2) is the catalytic component of the RNA-induced silencing complex (RISC) essential for RNA interference (RNAi).
- AGO2 binds short guide RNAs (miRNAs or siRNAs) to direct RISC to complementary mRNAs, leading to post-transcriptional gene silencing through endonucleolytic cleavage (slicing) or translational repression, depending on complementarity.
- It mediates translational inhibition by binding the mRNA cap, preventing eIF4E recruitment, and via interaction with EIF6, which blocks ribosomal subunit joining; targeted mRNAs accumulate in cytoplasmic P-bodies.
- AGO2 can also up-regulate translation of specific mRNAs (e.g., TNF) under serum starvation, and participates in transcriptional gene silencing (TGS) by targeting promoter regions with antigene RNAs.
