Biological Background: Argonaute-2 Function and Localization

  • Protein Argonaute-2 (AGO2, also known as eIF2C2) is the catalytic component of the RNA-induced silencing complex (RISC) essential for RNA interference (RNAi).
  • AGO2 binds short guide RNAs (miRNAs or siRNAs) to direct RISC to complementary mRNAs, leading to post-transcriptional gene silencing through endonucleolytic cleavage (slicing) or translational repression, depending on complementarity.
  • It mediates translational inhibition by binding the mRNA cap, preventing eIF4E recruitment, and via interaction with EIF6, which blocks ribosomal subunit joining; targeted mRNAs accumulate in cytoplasmic P-bodies.
  • AGO2 can also up-regulate translation of specific mRNAs (e.g., TNF) under serum starvation, and participates in transcriptional gene silencing (TGS) by targeting promoter regions with antigene RNAs.
  • During SARS-CoV-2 infection, AGO2 associates with viral miRNA-like small RNA CoV2-miR-O7a to repress host mRNAs such as BATF2, contributing to evasion of interferon response.
  • Subcellularly, AGO2 localizes to the cytoplasm (including P-bodies) and the nucleus.
  • Post-translational modifications include phosphorylation, hydroxylation, nitration, and ubiquitination; it requires magnesium or manganese for endonuclease activity.
  • Keywords highlight its roles in RNA-binding, ribonucleoprotein complex, host-virus interaction, and disease association (intellectual disability).

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide from a conserved region (amino acids 101-200), consistent with detection of human, mouse, and rat AGO2; however, always validate specificity in the relevant sample system.
  • For Western blot (WB), the expected band is ~97 kDa; include appropriate positive and negative controls and consider using a loading control for normalization.
  • For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimize antigen retrieval and antibody dilutions using known AGO2-expressing tissues or cells.
  • ELISA applications may require further optimization; consider checkerboard titrations to determine optimal coating and detection conditions.
