Biological Background: Arginase-1 Function and Localization
- Arginase-1 is a manganese-dependent hydrolase that catalyzes the conversion of L-arginine to urea and L-ornithine, a key reaction in the hepatic urea cycle for ammonia detoxification.
- Beyond the liver, it regulates L-arginine homeostasis in non-hepatic tissues, competing with nitric oxide synthase (NOS) for the shared substrate arginine, and thereby modulating nitric oxide production.
- In immune cells, notably alternatively activated M2 macrophages, arginase-1 supports wound healing, tissue regeneration, and immune suppression; its function is organ-specific and influences anti-parasitic and allergic responses. Related references: PMID:19360123, PMID:20483789, PMID:23552798
- Within the tumor microenvironment, it is expressed by infiltrating dendritic cells and myeloid-derived suppressor cells, where it dampens T cell-mediated antitumor immunity through arginine depletion. Related references: PMID:19414774, PMID:23248265
- In group 2 innate lymphoid cells (ILC2s), arginase-1 promotes acute type 2 lung inflammation and fuels optimal ILC2 proliferation, linking arginine metabolism to innate lymphoid cell function. Related references: PMID:27043409
- Subcellularly, it localizes to the cytoplasm and cytoplasmic granules, consistent with its role in rapid arginine metabolism upon release.
- Tissue expression is prominent in macrophages, ILC2s, and lung tumor-associated myeloid cells, reflecting its immunological and metabolic significance. Related references: PMID:12193690, PMID:19360123, PMID:7537672, PMID:27043409, PMID:15313928, PMID:19414774
Experimental Guidance and Technical Tips
- The immunogen is a recombinant mouse Arginase-1 protein; verify cross-reactivity with rat samples by sequence alignment, and consider using positive controls from mouse liver or macrophage lysates.
- For western blot (WB), expect a band at approximately 34 kDa; include negative controls (e.g., Arginase-1 knockout cell lines) to confirm specificity.
- In immunofluorescence (IF-P), optimize fixation (e.g., 4% PFA) and permeabilization due to the cytoplasmic localization; counterstain with a nuclear dye to distinguish compartments.
- For ELISA, pair this antibody with a suitable detection antibody after testing dynamic range and matrix effects in your sample type (e.g., tissue homogenates, cell lysates).
