Biological Background: Arginase-1 Function and Localization
- ARG1 is a key urea cycle enzyme in the liver, hydrolyzing L-arginine to urea and L-ornithine, which can be further metabolized to proline and polyamines that support collagen synthesis and cell proliferation.
- In nonhepatic tissues, ARG1 regulates L-arginine availability by competing with nitric oxide synthase (NOS), thereby modulating innate and adaptive immune responses.
- In polymorphonuclear granulocytes (PMNs), ARG1 participates in an antimicrobial effector pathway; upon PMN cell death, it is released and depletes arginine in the microenvironment, leading to suppressed T-cell and natural killer (NK) cell proliferation and cytokine secretion. - ARG1 promotes acute type 2 inflammation in the lung and supports optimal proliferation of group 2 innate lymphoid cells (ILC2s).
- Localized to the cytoplasm and cytoplasmic granules, ARG1 contains a manganese cofactor essential for its catalytic activity.
