Biological Background: ARF6 Function and Localization
- ARF6 is a small GTP-binding protein that cycles between inactive GDP-bound and active GTP-bound states, regulating membrane trafficking and actin cytoskeleton dynamics.
- It plays a critical role in endocytic recycling, sorting cargo from early endosomes to the plasma membrane, including palmitoylated proteins (PubMed:37461827).
Related references: PMID:37461827 - ARF6 is required for normal completion of mitotic cytokinesis, localizing to the cleavage furrow and midbody (By similarity).
- Involved in neuronal development: regulates dendritic spine development, dendritic branching, and filopodia extension (PubMed:14978216), and potentiates neurite outgrowth via interaction with APBB1 (PubMed:36250347).
Related references: PMID:14978216 PMID:36250347 - Functions in epithelial polarization and junctional remodeling by regulating surface levels of adherens junction proteins such as CDH1 (PubMed:36017701).
Related references: PMID:36017701 - During microbial infection, ARF6 acts as an allosteric activator of cholera toxin catalytic subunit and facilitates endocytosis of enterovirus 71 for viral entry into brain endothelial cells (PubMed:11266366, PubMed:16737952, PubMed:18400762).
Related references: PMID:11266366 PMID:16737952 PMID:18400762 - Subcellular localization includes cytoplasm, cell membrane, endosome membranes (especially recycling endosome), Golgi, filopodia, ruffles, and midbody. It is ubiquitously expressed with higher levels in heart, substantia nigra, and kidney.
- Keywords: GTP-binding, lipoprotein, myristate, protein transport, neurogenesis, cell division.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a central region (aa 76–175) of human ARF6, which shows high sequence identity across human, mouse, and rat, supporting the observed cross-reactivity.
- For Western blot, a band around 20 kDa is expected; consider using reducing conditions and appropriate loading controls.
- For ELISA, immobilize antigen or antibody according to standard protocols; specificity should be validated with recombinant ARF6 protein or cell lysates.
- As ARF6 is membrane-associated and may be palmitoylated, consider subcellular fractionation to enrich for membrane fractions if detection is weak in whole-cell lysates.
