Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1-180) of human ARF5. This region includes the conserved GTP-binding domain and is likely to detect full-length ARF5. Consider using a positive control lysate from HeLa, HEK293, or other human cell lines expressing ARF5 for Western blot validation.
- For Western blot, the predicted molecular weight of ARF5 is approximately 21 kDa. Use reducing SDS-PAGE conditions and a standard blocking buffer. Optimize primary antibody dilution empirically between 1:500 and 1:2000, and include a species-appropriate secondary antibody.
- The antibody is suitable for ELISA applications. For direct or sandwich ELISA, titrate the antibody against the recombinant protein used as immunogen or against cell lysates to determine optimal coating and detection concentrations.
