Biological Background: ARCN1 Function and Localization
- ARCN1 encodes coatomer subunit delta, also known as archain or delta-COP, a component of the coatomer (COPI) complex.
- The coatomer complex is a cytosolic protein complex that binds to dilysine motifs and reversibly associates with Golgi-derived non-clathrin-coated vesicles, mediating biosynthetic protein transport from the ER through the Golgi.
- Required for budding from Golgi membranes and essential for retrograde Golgi-to-ER transport of dilysine-tagged proteins.
- Recruitment to membranes is mediated by ADP-ribosylation factors (ARFs), small GTP-binding proteins.
- Influences Golgi structural integrity and also plays a role in the processing, activity, and endocytic recycling of LDL receptors (By similarity).
- Localizes to the cytoplasm, Golgi apparatus membrane, and COPI-coated vesicle membrane.
- Ubiquitously expressed in tissues.
- Post-translational modifications include acetylation and phosphorylation; undergoes alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen sequence spans amino acids 212–511 of human ARCN1, a region conserved in mouse and rat, consistent with reported cross-reactivity.
- For Western blotting, a band corresponding to approximately 57 kDa is expected; optimize loading and transfer conditions.
- In immunofluorescence/ICC, ARCN1 localizes to the Golgi apparatus; appropriate fixation and permeabilization may enhance detection.
- ELISA protocols: consider antigen coating concentration and antibody titration for optimal signal-to-noise ratio.
