Biological Background: ARAP1 Function and Localization
- ARAP1, also known as centaurin-delta-2 (Cnt-d2), is an Arf-GAP with Rho-GAP domain, ANK repeat and PH domain-containing protein 1 that modulates actin cytoskeleton remodeling.
- It acts as a phosphatidylinositol 3,4,5-trisphosphate (PIP3)-dependent GTPase-activating protein for ARF and RHO family GTPases, with a preference for ARF1 and ARF5. Related references: PMID:11804590, PMID:19666464
- ARAP1 positively regulates the ring size of circular dorsal ruffles and promotes macropinocytosis. Related references: PMID:22573888

- It is widely expressed in tissues including heart, skeletal muscle, spleen, kidney, liver, placenta, lung, peripheral blood leukocytes, adrenal gland, bone marrow, brain, lymph node, mammary gland, prostate, spinal cord, stomach, thyroid, and trachea.
- ARAP1 is a metal-binding phosphoprotein with zinc-finger domains, and undergoes alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a recombinant fragment spanning amino acids 834–1133 of human ARAP1, located in the C-terminal region. This antibody may recognize full-length ARAP1 and possibly N-terminal isoforms or fragments containing the epitope region.
- For Western blotting, the predicted molecular weight is approximately 162 kDa. Consider using a low-percentage SDS-PAGE gel (e.g., 6–8%) and appropriate molecular weight markers to resolve high-molecular-weight proteins.
- For immunofluorescence/immunocytochemistry, ARAP1 displays a multi-compartment localization (Golgi, endosomes, plasma membrane ruffles, podosomes); therefore, co-staining with organelle markers is recommended to confirm specific signals.
- The antibody shows cross-reactivity with mouse and rat ARAP1, based on sequence conservation, making it suitable for rodent model studies. Validate dilution and conditions in the relevant sample system.
- For ELISA, this polyclonal antibody can serve as a capture or detection reagent; optimization of coating and detection parameters is advised.
