Biological Background: ARAF Function and Localization

  • Serine/threonine-protein kinase that belongs to the RAF family of proto-oncogenes. Encoded by the ARAF gene, with alternative splicing producing multiple isoforms.
  • Key component of the RAS-RAF-MEK-ERK signal transduction pathway, relaying mitogenic signals from the cell membrane to the nucleus to regulate cell proliferation and differentiation.
  • Also implicated in the TOR signaling cascade, linking growth factor signals to cellular metabolism and growth.
  • Phosphorylates PFKFB2, a bifunctional enzyme that controls glycolysis, providing a direct link between ARAF activity and metabolic regulation. Related references: PMID:36402789
  • Functions as a positive regulator of myogenic differentiation by inducing cell cycle arrest, upregulating myogenin and other muscle-specific proteins, and promoting myotube formation.
