Biological Background: Aquaporin-8 (AQP8) Function and Localization
- Aquaporin-8 (AQP8) is a channel protein that facilitates the permeation of water and uncharged molecules, such as hydrogen peroxide and neutral ammonia (NH3), across cellular membranes including plasma membrane, inner mitochondrial membrane, and endoplasmic reticulum membrane. Related references: PMID:15948717, PMID:18948439, PMID:23541115
- At the canalicular plasma membrane, AQP8 mediates osmotic water transport toward the bile canaliculus, facilitating cAMP-induced bile canalicular water secretion essential for bile formation. Related references: PMID:18948439
- AQP8 releases hydrogen peroxide from hepatocyte mitochondria, modulating SREBF2-mediated cholesterol synthesis, and facilitates mitochondrial ammonia uptake for urea production, mainly under glucagon stimulation. Related references: PMID:30579780, PMID:34292591
- In B cells, AQP8 transports CYBB-generated hydrogen peroxide from the outer leaflet of the plasma membrane into the cytosol to promote activation and differentiation. (By similarity)
- AQP8 is expressed in pancreas, colon, and sperm midpiece; it localizes to cell membrane, mitochondrial inner membrane, apical and basolateral membranes, and smooth endoplasmic reticulum. Related references: PMID:28042826
- The protein has a molecular weight of approximately 27 kDa and contains transmembrane helices, a repeat structure, and glycosylation sites.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 100-200 of human AQP8 (NP_001160.2). For western blot, a band around 27 kDa is expected; optimize antibody dilution and validate in your sample system.
- Suitable applications include WB, IF/ICC, and ELISA. For immunofluorescence, consider appropriate fixation and permeabilization protocols for mitochondrial or membrane targets.
- Cross-reactivity with mouse and rat is reported; validate in relevant tissue samples such as pancreas, colon, or liver.
- Note that the AQP8 water transport mechanism is mercury- and copper-sensitive; if studying function, include appropriate controls.
