Biological Background: APP Function and Localization

  • Amyloid-beta precursor protein (APP) is a transmembrane glycoprotein with several isoforms generated by alternative splicing; the major neuronal isoform is APP695, while APP751 and APP770 are expressed in non-neuronal cells, and appican is expressed in astrocytes.
  • APP functions as a cell surface receptor involved in neurite growth, neuronal adhesion, and axonogenesis; interactions between APP molecules on adjacent cells promote synaptogenesis.
    Related references: PMID:25122912
  • APP serves as a kinesin I membrane receptor, mediating axonal transport of beta-secretase and presenilin 1, which is crucial for cargo delivery towards synapses.
    Related references: PMID:17062754, PMID:23011729
  • The amyloid-beta peptides, derived from APP proteolysis, are lipophilic metal chelators that bind copper, zinc, and iron, and can reduce Cu²⁺ and Fe³⁺; they promote tau aggregation and TPK II-mediated phosphorylation, and their interaction with overexpressed HADH2 contributes to oxidative stress.
  • Gamma-CTF peptides and caspase-cleaved fragments, such as C31, are potent enhancers of neuronal apoptosis.
  • APP contains a Kunitz-type serine protease inhibitor (BPTI) domain in some isoforms, conferring protease inhibitor activity, and undergoes extensive post-translational modifications including N- and O-glycosylation, phosphorylation, and sulfation.
  • Subcellularly, APP is localized to the cell membrane, clathrin-coated pits, early endosomes, endoplasmic reticulum, Golgi apparatus, cytoplasmic vesicles, and is also secreted; it can be detected in the nucleus and cytoplasm.
  • APP is expressed in the brain and cerebrospinal fluid, with highest levels in the frontal lobe of the cortex; it is also found in kidney, heart, spleen, and plasma. Isoform APP695 predominates in neurons, while APP751 and APP770 are expressed in non-neuronal cells (PubMed:2649245).

Experimental Guidance and Technical Tips

  • Due to the immunogen design (amino acids 1-100 of human Aβ42), the antibody is expected to recognize both full-length APP (~87 kDa) and amyloid-beta-containing proteolytic fragments, such as CTF99, CTF83, and Aβ peptides, in Western blot. Validation in wild-type versus APP knockout samples may help confirm specificity, given the KO-validated status.
  • For ELISA, consider pairing this polyclonal antibody with a monoclonal antibody recognizing a different APP epitope to develop a sensitive sandwich assay. Optimization of coating and detection conditions is recommended.
  • Although the antibody is raised against human APP and shows cross-reactivity with human samples, users working with other species should conduct appropriate validation due to potential sequence divergence.
  • The antibody is shipped on ice bags to maintain stability; upon receipt, aliquot and store at -20°C or -80°C to avoid repeated freeze-thaw cycles.
